Simultaneous gene expression and multi-gene silencing in Zea mays using maize dwarf mosaic virus

چکیده

Abstract Background Maize dwarf mosaic virus (MDMV), a member of the genus Potyvirus , infects maize and is non-persistently transmitted by aphids. Several plant viruses have been developed as tools for gene expression silencing in plants. The capacity MDMV both were examined. Results Infectious clones an Ohio isolate MDMV, OH5, obtained, engineered only, simultaneous marker virus-induced (VIGS) three endogenous target genes. Single single insertion constructs green fluorescent protein (GFP) genes double construct was demonstrated. Constructs with GFP inserted N-terminus HCPro more stable than those at CP our study. Unexpectedly, two sites also retained insertions higher rate single-insertion constructs. Engineered VIGS transmissible aphids ( Rhopalosiphum padi ). Conclusions These results demonstrate that MDMV-based vector can be used tool multi-gene maize.

Sugarcane Mosaic Virus-Based Gene Silencing in Nicotiana benthamiana

Background:Potyvirus-based virus-induced gene silencing (VIGS) is used for knocking down the expression of a target gene in numerous plant species. Sugarcane mosaic virus (SCMV) is a monopartite, positive single strand RNA virus. Objectives:pBINTRA6 vector was modified by inserting a gene segment of SCMV in place of Tobacco rattle virus (TRV) genome part 1 (TRV1 or RNA1)...

Cloning and Expression Analysis of ZmERD3 Gene From Zea mays

Background: Stresses (such as drought, salt, viruses, and others) seriously affect plant productivity. To cope with these threats, plants express a large number of genes, including several members of ERD (early responsive to dehydration) genes to synthesize and assemble adaptive molecules. But, the function of ERD3 gene hasn’t been known so far.Objectives:</strong...

Tomato and Tobacco Phytoene Desaturase Gene Silencing by Virus-Induced Gene Silencing (VIGS) Technique

Background and Aims: Virus-Induced Gene Silencing (VIGS) is a virus vector technology that exploits antiviral defense mechanism. By infecting plants with recombinant viruses containing host genes inserted in the viral genome, VIGS achieves the RNA silencing process. The purpose of this study was to investigate the possibility of tomato (Lycopersicon esculentum Mill.) and tobacco (Nicotiana be...

Phytosulphokine gene regulation during maize (Zea mays L.) reproduction.

The sulphated pentapeptide phytosulphokine (PSK) was identified as a substance that promotes cell division in low-density suspension cultures and has been implicated in various aspects of tissue differentiation in plants. The peptide is derived from PSK precursor proteins that are encoded by small gene families. The physiological roles of PSK are still not clearly defined and little is known ab...

Genome-wide identification, splicing, and expression analysis of the myosin gene family in maize (Zea mays)

The actin-based myosin system is essential for the organization and dynamics of the endomembrane system and transport network in plant cells. Plants harbour two unique myosin groups, class VIII and class XI, and the latter is structurally and functionally analogous to the animal and fungal class V myosin. Little is known about myosins in grass, even though grass includes several agronomically i...
